The DeepCatch™ Broad-Range Nucleic Acid Concentration Kit is designed to rapidly concentrate purified DNA and RNA into ultra-low volumes while providing high recovery across a broad size range, including ultra-short fragments down to 10 nt. Developed for downstream assays where target nucleic acids are extremely dilute, this carrier-free system maximizes the final sample concentration without introducing exogenous nucleic acid interference.

Low-abundance cfDNA, cfRNA, and miRNAs are often recovered in volumes that exceed the input limits of NGS, ddPCR, qPCR, and RT-qPCR. As a result, only a portion of the available nucleic acid may enter each reaction.
Conventional concentration methods may lose ultra-short fragments, co-concentrate salts and inhibitors, or require larger elution volumes. Verified Retention of Trace 10 nt fragments

A 10 ng FAM-labeled 10 nt oligonucleotide was diluted in 200 µL of low TE buffer and concentrated to a final volume of 5 µL using the DeepCatch™ Broad-Range Nucleic Acid Concentration Kit. Lane 1: 10 ng input; Lane 2: a 5 µL aliquot of the 200 µL sample before concentration; Lane 3: the entire 5 µL concentrated eluate. The concentrated eluate showed band intensity visually comparable to the input, demonstrating strong retention of the 10 nt oligonucleotide during a 40-fold reduction in sample volume. A faint, faster-migrating band in the input, consistent with free FAM dye, was not detected in the concentrated eluate, indicating that this low-molecular-weight non-nucleic-acid impurity was not co-concentrated under the tested conditions. Samples were analyzed by 15% denaturing urea-PAGE.

A nucleic acid mixture containing 10, 15, 20, and 90 nt fragments was diluted in 200 µL of TE buffer and concentrated to 5 µL using the DeepCatch™ Broad-Range Nucleic Acid Concentration Kit. Lane 1: input mixture; Lane 2: a 5 µL aliquot of the 200 µL sample before concentration; Lane 3: the entire 5 µL concentrated eluate. Samples were analyzed by 15% denaturing urea-PAGE followed by SYBR Green staining. All tested fragment sizes, including the ultra-short 10 nt fragment, were readily detected in the concentrated eluate, demonstrating broad-range retention with no obvious size-dependent loss during a 40-fold reduction in sample volume.
